Anyone else here play Mass Effect? That could be a really good theme. Lots of characters to choose from for the roles.

I vote for Portal! It can follow the last game.

SPOILERS

GlaDOS finds that, even with the many frozen people found, none of the subjects satisfy the "itch" the way Chell did. The new subjects are incapable of completing the tests, and so no useful data can be collected.

In addition, the emotion that Chell brought about has been plaguing GlaDOS ever since she left. Why? Why did this happen? What has changed?

Wanting to gather more data and answers, GlaDOS captures Chell again for one final experiment. Every day and night, another test.

The other players could be either the newly-thawed human subjects, with the Mafia being either those driven paranoid and seeking to be the last ones standing, or double agents secretly working for GlaDOS.

Or, a better idea, the other players could be the robots (P-body, Atlas, the cores, etc.), with Mafia likely being agents for GlaDOS.

Why GlaDOS would want Chell and the others dead, however, someone else can think up.

Both of these themes would work out well, I feel. Though I think that you'd have a better time doing Phoenix Wright, since you know the games a lot better, right?
